That which does not kill yeast makes it stronger

ScienceDaily (Jan. 29, 2012) ? Cells trying to keep pace with constantly changing environmental conditions need to strike a fine balance between maintaining their genomic integrity and allowing enough genetic flexibility to adapt to inhospitable conditions. In their latest study, researchers at the Stowers Institute for Medical Research were able to show that under stressful conditions yeast genomes become unstable, readily acquiring or losing whole chromosomes to enable rapid adaption.

The research, published in the January 29, 2012, advance online issue of Nature, demonstrates that stress itself can increase the pace of evolution by increasing the rate of chromosomal instability or aneuploidy. The observation of stress-induced chromosome instability casts the molecular mechanisms driving cellular evolution into a new perspective and may help explain how cancer cells elude the body's natural defense mechanisms or the toxic effects of chemotherapy drugs.

"Cells employ intricate control mechanisms to maintain genomic stability and prevent abnormal chromosome numbers," says the study's leader, Stowers investigator Rong Li, Ph.D. "We found that under stress cellular mechanisms ensuring chromosome transmission fidelity are relaxed to allow the emergence of progeny cells with diverse aneuploid chromosome numbers, producing a population with large genetic variation."

Known as adaptive genetic change, the concept of stress-induced genetic variation first emerged in bacteria and departs from a long-held basic tenet of evolutionary theory, which holds that genetic diversity -- evolution's raw material from which natural selection picks the best choice under any given circumstance -- arises independently of hostile environmental conditions.

"From an evolutionary standpoint it is a very interesting finding," says graduate student and first author Guangbo Chen. "It shows how stress itself can help cells adapt to stress by inducing chromosomal instability."

Aneuploidy is most often associated with cancer and developmental defects and has recently been shown to reduce cellular fitness. Yet, an abnormal number of chromosomes is not necessarily a bad thing. Many wild yeast strains and their commercial cousins used to make bread or brew beer have adapted to their living environs by rejiggering the number of chromosomes they carry. "Euploid cells are optimized to thrive under 'normal' conditions," says Li. "In stressful environments aneuploid cells can quickly gain the upper hand when it comes to finding creative solutions to roadblocks they encounter in their environment."

After Li and her team had shown in an earlier Nature study that aneuploidy can confer a growth advantage on cells when they are exposed to many different types of stress conditions, the Stowers researchers wondered whether stress itself could increase the chromosome segregation error rate.

To find out, Chen exposed yeast cells to different chemicals that induce various types of general stress and assessed the loss of an artificial chromosome. This initial screen revealed that many stress conditions, including oxidative stress, increased the rate of chromosome loss ten to 20-fold, a rate typically observed when cells are treated with benomyl, a microtubule inhibitor that directly affects chromosome segregation.

The real surprise was radicicol, a drug that induces proteotoxic stress by inhibiting a chaperone protein, recalls Chen. "Even at a concentration that barely slows down growth, radicicol induced extremely high levels of chromosome instability within a very short period of time," he says.

Continued growth of yeast cells in the presence of radicicol led to the emergence of drug-resistant colonies that had acquired an additional copy of chromosome XV. Yeast cells pretreated briefly with radicicol to induce genomic instability also adapted more efficiently to the presence of other drugs including fluconazole, tunicamycin, or benomyl, when compared to euploid cells.

Interestingly, certain chromosome combinations dominated in colonies that were resistant to a specific drug. Fluconazole-resistant colonies typically gained an extra copy of chromosome VIII, tunicamycin-resistant colonies tended to lose chromosome XVI, while a majority of benomyl-resistant colonies got rid of chromosome XII. "This suggested to us that specific karyotypes are associated with resistance to certain drugs," says Chen.

Digging deeper, Chen grew tunicamycin-resistant yeast cells, which had adapted to the presence of the antibiotic by losing one copy of chromosome XVI, under drug-free conditions. Before long, colonies of two distinct sizes emerged. He quickly discovered that the faster growing colonies had regained the missing chromosome. By returning to a normal chromosome XVI number, these newly arisen euploid cells had acquired a distinctive growth advantage over their aneuploid neighbors. But most importantly, the fast growing yeast cells were no longer resistant to tunicamycin and thus clearly linking tunicamycin resistance to the loss of chromosome XVI.

Researchers who also contributed to the work include William D. Bradford and Chris W. Seidel both at the Stowers Institute for Medical Research.

Pardoned by Haley Barbour, a 'free man' is on the run

Convicted for killing a convenience store clerk in 1994, Joseph Ozment walked out of the Governor's Mansion after being pardoned by Gov. Haley Barbour on Jan. 8 and hasn't been seen since.

Declaring Joseph Ozment ?rehabilitated,? Gov. Haley Barbour included the convicted killer among over 200 pardons he issued in his last days as governor of Mississippi.

Mr. Ozment was last seen leaving the Governor's Mansion, where he was a convict ?trusty,? on Jan. 8 when he got into a car driven by his grandmother.

Ozment, whom Barbour described Friday as a ?free man,? is now being sought by Mississippi authorities investigating the constitutionality of Barbour's mass pardons, which shocked many Mississippians, including victims and law enforcement. The list included over 40 murderers, rapists and others convicted of violent crimes.

The unusual manhunt is the latest twist in a peculiar tale of Southern patriarchy and redemption that has dogged Mr. Barbour since he left office earlier this month. The governor has defended his actions, saying the state pardon board had already freed most of the people, and that the clemency was mainly designed to give worthy ex-convicts the right to vote and hunt.

But national scrutiny has revealed that those pardoned were both disproportionately white and many had access to powerful interests in the state. In the aftermath, the state ended its mansion ?trusty? program, a judge is deciding the constitutionality of the majority, and the legislature is weighing several bills to curtail the pardon process.

At the same time, the pardons also touched on deeper issues around the nature of redemption and mercy for a country that has one of the highest incarceration rates in the world.

?While his timing and transparency are in question, he has at least reopened a needed national discussion on how justice must be tempered by mercy,? wrote the Monitor's editorial board last week.

But in Mississippi, that debate has taken backseat to concerns about Ozment's whereabouts. While four other former mansion ?trusties? that were released have checked back in with the judge, and vowed to maintain daily contact, Ozment has disappeared.

Attorney General Jim Hood, who called Barbour's mass pardons ?a slap to the face? of victims and the judicial system, said Ozment was last seen in northwest Mississippi, from where he hails.

A CNN crew trying to track him down also traveled to Memphis, Tenn., and Birmingham, Ala., where Ozment has family, but have so far come up short. On Jan. 17, the network carried an interview with Anthony McCray, one of the five former mansion ?trusties? pardoned by Barbour. Mr. McCray called Ozment and the other trusties ?nice guys,? and suggested that ?God touched Haley Barbour's heart? as the reason why Barbour signed the pardons.

The search has raised unprecedented issues, including the extent to which the state can legally force Ozment, who's not wanted for any crime and now has a clean criminal record, to report to a judge.

The issue became only murkier on Friday when Attorney General Jim Hood hinted that there could be a financial reward for information on his whereabouts. Mr. Hood has said he may begin a criminal investigation if Ozment continues to refuse to surface.

The fact that Ozment hasn't abided by a judge's order to report for a hearing suggests to Hood that he may be a threat to public safety.

?He doesn't have a lot to lose if he thinks he's going back to prison for life," Hood said Friday. "That's what concerns me about the public safety."

3M posts small profit increase in 4Q (AP)

NEW YORK ? 3M says its profit inched 3 percent higher in the fourth-quarter as growth in basic products like Post-Its and police tape offset declines in its high-tech products.

The maker of everything from Scotch Tape to computer arms also maintained its profit forecast for 2012 as it expects slower economic growth around the world.

The Maplewood, Minn., company said Thursday it earned $954 million, or $1.35 per share, compared with $928 million, or $1.28 per share, a year ago.

Revenue rose 6 percent to $7.09 billion.

FactSet says analysts expected a profit of $1.31 per share on revenue of $7.09 billion.

3M Co. said sales were strongest for the company's industrial and transportation unit. Sales fell in its electronics and communications and display and graphics segments.

Humans' Taste for Dolphins & Manatees on the Rise (LiveScience.com)

Fillet of dolphin? Polar bear steak? As world population increases, people in coastal poverty-stricken areas are turning to the ocean for their meals, consuming marine mammals such as dolphins and seals, new research suggests.

Since 1990, at least 87 species of marine mammals ? including dolphins, porpoises and manatees ? have been served up in 114 countries. They are the victims of hunting and even commercial fishing operations, where they are sometimes caught accidentally, the researchers said.

The fishing of larger marine mammals, like humpback whales, is strictly regulated and monitored; but the extent to which these smaller warm-blooded marine species, including dolphins and seals, are caught, killed and eaten has been largely unstudied and unmonitored.

"International regulatory bodies exist to gauge the status of whale populations and regulate the hunting of these giants," study researcher Martin Robards, of the Wildlife Conservation Society, said in a statement. "These species, however, represent only a fraction of the world's diversity of marine mammals, many of which are being accidentally netted, trapped, and ? in some instances ? directly hunted without any means of tracking as to whether these off-takes are sustainable."

Porpoise and narwhal on the menu

To get a clearer picture of the problem, the Wildlife Conservation Society and Okapi Wildlife Associates examined records on small fisheries focused on small whales (like pilot whales), dolphins and porpoises from 1975 and records of global marine mammal catches between 1966 and 1975.

From there, the researchers consulted about 900 other sources, including reports and discussions with numerous researchers and environmental managers; the exhaustive investigation took three years to complete.

They found that since 1990, people in at least 114 countries have consumed one or more of at least 87 marine mammal species.The list includes species people might not know by name or sight, such as the pygmy beaked whale, South Asian river dolphin, narwhal, Chilean dolphin, long-finned pilot whale and Burmeister's porpoise. The list also includes well-known species, such as bottlenose dolphins, seals, sea lions (including the California sea lion), polar bears and three species of manatees. Some of these species, like the manatee's close relative the dugong, are considered a delicacy in some parts of the world, making them targets of human consumption.

Wild eats

Since the 1970s, humans' taste for these warm-blooded aquatic animals has apparently been on the rise, the researchers found, especially incoastal areas and estuaries (where rivers meet oceans). This could be due, in part, to changes in fishing techniques in those areas, where these marine mammals are caught as "bycatch" in nets meant for other fish.

In areas such as the Congo, Gabon and Madagascar, these marine mammals serve as supplementary sources of dietary protein, similar to the animals in the forests that are taken by hunters and locals as bushmeat. As the world's population continues to increase, so does its food needs. The Wildlife Conservation Society is working with fishermen in these areas to reduce the need to catch wild marine mammals, and instead hunt sustainable fish.

The researchers say that increased awareness of the problem and increased monitoring are needed to prevent the destruction of marine life.

"There is a need for improved monitoring of species such as Atlantic and Indo-Pacific humpback dolphins," Howard Rosenbaum, director of the Wildlife Conservation Society's Ocean Giants Program, said in a statement. "In more remote areas and a number of countries, a greater immediate need is to understand the motivations behind the consumption of marine mammals and use these insights to develop solutions to protect these iconic species."

The study was published Jan. 24 in the journal Biological Conservation.

France's Hollande raises election bar for Sarkozy (Reuters)

PARIS (Reuters) ? Socialist presidential challenger Francois Hollande has raised the bar with a weighty economic plan and nimble performance in a televised debate this week, and President Nicolas Sarkozy is preparing to fight back with a TV interview on Sunday.

With Sarkozy aiming to dazzle voters with crisis measures to stem rampant unemployment, Hollande laid out plans on Thursday to squeeze tax breaks on the wealthy and big companies to fund investment in small businesses, education and new jobs.

Keeping new tax revenues carefully higher than spending, the program re-established Hollande as a fiscally responsible centre-leftist, after his recent attack on the financial sector, even if some economists found it lacking in structural reform ideas.

It also catapulted him ahead of Sarkozy -- who, to the annoyance of some in his conservative UMP party, may not announce his re-election bid until early March -- in terms of laying out all the detail of his campaign pledges.

Hollande, who tops opinion polls but had been criticized for a lack of campaign direction, also ably fended off barbs in a primetime TV duel with Alain Juppe, Sarkozy's foreign minister and one of the most experienced politicians on his team.

"This is a key moment for (Sarkozy) because it comes after a week that has been a very good one for Hollande," said Brice Teinturier, deputy director at pollster Ipsos.

"Hollande has clearly retaken the offensive and above all he has disproved two of the UMP's criticisms of him: a lack of presidential character and weight, and having flabby proposals, because he's come up with 60 that are pretty precise."

Often seen as a weaker public speaker than the flamboyant and hot-blooded Sarkozy, the softer-spoken Hollande took the upper hand in a long and highly technical debate against Juppe, a former prime minister under President Jacques Chirac.

Hollande proved unflappable and eloquent -- and even looked like he was rather enjoying himself -- as Juppe sniped at his ideas and accused him of being arrogant and vague.

The long-time Socialist party boss managed to get in the last word on most exchanges.

ACHIEVABLE GOALS

Sarkozy is planning a last-minute campaign where he will admit that the global economic crisis derailed some of his initial promises and will try to show that he is the safest pair of hands to pull France out of the latest bout of turmoil.

Jolted by a cut to France's AAA credit rating by Standard and Poor's this month, Sarkozy has switched his focus to growth and will use Sunday's panel interview to flesh out a set of job-saving measures agreed in hurried talks with unions on January 18.

Party officials say his campaign will centre on the need to restore competitiveness, which has slid far below Germany's over the past few years as France raised wages and shortened the work week while Germany kept wages low and job contracts flexible.

Aides say Sunday's 1900 GMT interview will reflect that.

"He will promise things that are achievable," one of Sarkozy's long-term advisers told Reuters. "His obsession now is economic competitiveness. When you are trying for re-election you can't promise 50 measures: you have to say 'I am a better bet than the other guy' and be much more focused."

The economy and the euro zone crisis are the top concerns for voters ahead of the two-round April 22 and May 6 ballot.

Left-wingers have accused Sarkozy of overblowing the scale of the crisis to serve his political needs, while Sarkozy is playing up his experience in office and hoping the fact Hollande has never been a government minister will count against him.

Credibility on the economy will be key to winning over centrist voters, many of whom would have backed former finance minister and IMF head Dominique Strauss-Kahn for president, until his career was wrecked by a sex assault scandal in New York last May, and are still weighing Hollande's credentials.

A surge in support for centrist Francois Bayrou, who is scoring 14-15 percent in polls behind Hollande, Sarkozy and far-right leader Marine Le Pen, shows how key centre-ground votes will be in a runoff likely to pit Hollande against Sarkozy.

Bayrou has been critical of Hollande's ideas, and may find it hard to back him in a runoff after Hollande declared war on the world of finance in a speech last weekend. Hollande said on Thursday he did not see a job for Bayrou in his government.

Tax Returns and Tithing: How Mitt Romney Gives Away 16% of His Income (Time.com)

When Mitt Romney released his 2010 tax returns on Tuesday, the one number that probably stood out to many Americans wasn't his 14% effective tax rate or his $20 million-plus annual income. It was the $7 million he gave to charity over the last two years, including some $4.1 million to the Church of Jesus Christ of Latter-Day Saints.

Where does that money go? In addition to donating to his family's Tyler Foundation, Romney does his duty as an active LDS member. The Mormon Church requires its members to tithe 10% of their income, and Romney's contributions match that responsibility. (PHOTOS: The Rich History of Mitt Romney)

Designed to follow the Biblical mandate to feed the hungry and clothe the naked, the Mormon tithing system supports a giant welfare infrastructure. In addition to financing temple construction and missionary programs, tithing supports more than 300 employment resource centers and 80 family services offices around the world. The church employs some 8,500 missionaries who teach English, give agricultural aid, provide medical practices and distribute clothing. It even stores a 3-6 month food supply so its members won't go hungry in the event of a disaster, and most families forgo two consecutive meals a month to give money as a "fast offering" for the poor. Local bishops -- a position Romney once held -- work with members of their local church wards to overcome economic hardships, and are even empowered to pay a family's mortgage in the hardest of times.

Structured or not, these donations set Romney apart from his political colleagues. In 2010, Romney gave away 16% of his income while Newt Gingrich's returns show he gave only 2.6%. The portion Gingrich donated to his Washington D.C. home church, the Basilica of the National Shrine of the Immaculate Conception, was even smaller -- 0.3%, or $9,540. Because the Gingrichs' also reported an earned income of $5,918 from the National Shrine -- Callista sings as an alto in the church's professional choir, which pays $80 per mass and rehearsal -- so the net balance of their contributions sinks below $4,000. The rest of Gingrich's charitable donations went to unspecified cash contributions through the Gingrichs' businesses, some $68,500, and to miscellaneous donations, near $3,100. The Obamas, meanwhile, gave 14% of their income to a total of 36 different charities in 2010. Much of that went to the Fisher House Foundation, a charity that works with veterans, and smaller amounts went to the Clinton Bush Haiti Fund and the Boys and Girls Club of America. In Monday's debate, Romney said he is "proud of the fact that I pay a lot of taxes." Though he's at times reluctant to speak publicly about his Mormon faith, his charitable giving, half of which goes to the socially-active LDS church, is something to be proud of as well.

Jet-setting actor/musician Joe Jonas has once again made time in his busy schedule for a little stop through Cleveland — "Hot in Cleveland," that is, the popular show on TV Land starring Valerie Bertinelli, Betty White, Wendie Malick and Jane Leeves.

Jonas has a guest-starring role on the sitcom as the wayward son of Bertinelli's character. MTV News was lucky enough to visit the set of "Cleveland" on the day Jonas filmed his second guest spot, which premieres Wednesday night (January 25) at 10 p.m. ET. When we caught up with him for a few minutes between scenes, he shared some details about his return to the show.

"Being back on the show, [my character Will is] engaged, and I have to be engaged to Jennifer Love Hewitt, which is really nice," he said. "We're both sons and daughters of characters on the show, and we show up and surprise everybody. They don't expect us to be engaged."

Jonas said that while it was fun to work with Hewitt, their characters' engagement doesn't exactly last for long. "Both of our characters are really different. I think I'm a little bit blinded by the fact that she may not be the perfect person for me. I think we last probably less than Kim Kardashian in this," he joked.

Long engagements or no, Jonas said he loves his time on the show and would be happy to come back any time.

"I would love to be able to return again. The first episode when I left, people were like, 'Are you going to be back soon?' and I said, 'I think so.' I don't think my character passed away, so I think we're good. I don't die in this episode either," he added. "So I'm still around, so I could definitely come back if they'd like me to. I have a fun time being on the set and I love the cast; it's kind of like a family."

France tempers talk of quick Afghanistan pullout

(AP) ? France's troops won't be pulled hastily from Afghanistan, says foreign minister Alain Juppe, adding that talk of a retreat by the end of 2012 isn't "well thought out and examined."

Speaking Tuesday during a question and answer session in parliament, Juppe said "when I hear talk of an immediate pullout, or even by the end of 2012, I'm not sure that's well thought out and examined."

Last week Francois Hollande ? the Socialist front-runner in next spring's presidential election ? pledged to bring France's roughly 4,000 troops home from Afghanistan by the end of the year.

That followed President Nicolas Sarkozy's comment that if security for troops is not restored, "then the question of an early withdrawal of the French army would arise."

Sarkozy suspended training missions and threatened to withdraw French troops early from the NATO-led mission in Afghanistan following Friday's shooting deaths of four French troops by an Afghan soldier.

A team of police investigators will travel to eastern Afghanistan to investigate the killings at a base in eastern Gwan, according to a French military official.

Five or six investigators are to travel Wednesday or Thursday in response to a request for help from authorities who are currently holding a suspect and have asked for support from French police.

The official spoke Tuesday on condition of anonymity because of the sensitivity of the matter.

French Defense Minister Gerard Longuet traveled to Afghanistan to investigate over the weekend and is to issue a report to Sarkozy.

Current plans are for French troops to return home by 2014, when NATO is due to wind up its combat mission in Afghanistan.

In Afghanistan Tuesday, President Hamid Karzai declared an emergency in the mountainous Badakhshan province in the northeast.

He promised a relief fund of $160,000 after heavy snow and avalanches killed at least 46 people in the last week. Avalanches are an annual problem in the mountainous country.

Boy meets girl, grandpa meets robot in quirky Sundance comedies (Reuters)

PARK CITY, Utah, Jan 22 (TheWrap.com) ? Saturday's substantial snowfall may have slowed the Park City shuttle buses that take Sundance audiences from screening to screening, but the film festival kept on keeping on, with two afternoon world premieres that -- like previous Sundance hits "Little Miss Sunshine" and "The Kids Are All Right" -- tweak genre conventions while remaining exceedingly audience-friendly.

Frank Langella gives another compelling performance in "Robot and Frank," a movie that often felt like a cross between "Driving Miss Daisy" and the senior-citizens-rob-a-bank comedy "Going in Style," with a science fiction twist.

Set in the near future (you can tell because the adults all have names like "Hunter" and "Madison"), the film sees the elderly Frank (Langella), who's fighting off dementia, being left in the care of a nurse robot (voiced by Peter Sarsgaard) provided to Frank by his estranged son (James Marsden). We come to realize that Frank used to be a cat burglar, and since the robot hasn't been programmed with a moral compass, Frank teaches him how to pick locks and to be the perfect criminal sidekick.

The pensioner's health blossoms now that he has a new project to tackle, but what if he gets caught? What if his hippie-dippie, anti-robot daughter (Liv Tyler) moves back in to take care of him? And what about Frank's romance with the sexy local librarian (Susan Sarandon)?

"Robot and Frank" often winds up being the sum of its gimmick, but this talented cast (assembled by first-time director Jake Schreier) makes the movie loads of fun. Screenwriter Christopher D. Ford, also making his debut, even tosses in a few surprising third-act twists, and when's the last time you didn't see one of those coming?

The dialogue of teen rom-com "The First Time" winds up being one of the film's big flaws, but the cast and storytelling is so charming that you forgive the fact that almost all of the characters talk like screenwriters. (In this case, Jonathan Kasdan, who also directed.)

Kasdan's TV credits include "Dawson's Creek" and "Freaks and Geeks," but you'd never guess it from the quippy, artificial banter that takes up so much of this often delightful story about two high-schoolers who meet, fall hard and have sex over the course of one weekend. The sex part, of course, winds up complicating matters more than they ever could have expected, and the two have to figure out if they have a future together after that first awkward naked encounter.

It's hard not to be won over by "The First Time," though, since TV vets Britt Robertson ("The Secret Circle") and Dylan O'Brien ("Teen Wolf") are so utterly charming. Robertson, in particular, brings a brainy-pixie vibe that reminded me of the late, great Adrienne Shelley in those early Hal Hartley movies.

As for O'Brien, he's certainly charismatic, but he's tough to buy as the nerdy sensitive guy (who another character actually calls "average-looking") when he could clearly be an underwear model.

Nonetheless, the two actors click as slightly eccentric adolescents who feel like real people, even if they don't always talk like them.

Still, even if Kasdan bobbles the dialogue, he gets a lot of high school life just right, from messy keggers to Saturday night at the multiplex to the difference between calling a girl on her cell and calling her parents' number. Teenagers are probably likely to be way more forgiving of the beyond-their-years sophistication of the witty chat, and they might even make "The First Time" a hit.

Dentist accused of paper clip use in root canals

(AP) ? A former dentist in Massachusetts has pleaded guilty to Medicaid fraud for using paper clips instead of stainless steel posts in root canals.

Michael Clair is scheduled to be sentenced next Monday after pleading guilty last week in New Bedford Superior Court to a variety of charges, including defrauding Medicaid of $130,000 assault and battery, illegally prescribing prescription drugs and witness intimidation.

Prosecutors say the 53-year-old Clair was suspended by Medicaid in 2002, but continued to file claims from August 2003 to June 2005 by using the names of other dentists in his Fall River practice.

Authorities say instead of stainless steel posts for root canals, he used sections of paper clips ? which can cause pain and even infection ? in an effort to save money.

Euronet reports breach at European business (Reuters)

(Reuters) ? Payment processor Euronet Worldwide Inc said a "small portion" of its European business was the target of a criminal security breach late last year, sending its shares down as much 6 percent.

The hacking of the system, which was revealed on Monday through a regulatory filing, affected a part of Euronet's processing business that accounts for less than 5 percent of its revenue, profit and transactions, the company said.

"It was brought to our attention by the card organizations," Euronet Chief Executive Michael Brown told Reuters by phone.

"When we heard the first little inklings of this, we jumped in, figured it out, got third parties involved who are real experts at this, and closed the breach... between our discovery and our shutdown, it wasn't a long period of time."

About the delay in disclosing the incident, Brown said it was not a severe breach.

"Maybe the reason a lot of people didn't make a big deal about it, is just because the severity wasn't as bad as some other people have seen...We had a limited amount of bad activity and we've been free and clear for over a month," Brown said.

Expenses from the breach were less than 1 cent per share in the fourth quarter of 2011.

No claims or losses were asserted against the company in the fourth quarter 2011, Kansas-based Euronet said.

The company said it was working closely with international card associations and law enforcement agencies, but declined to comment if customers' personal data was stolen in the breach.

Several companies including Citigroup, Sony's Playstation network and Alliance Data's Epsilon unit were targets of hacking attacks last year.

Shares of the company closed at $18.23 on Monday on the Nasdaq.

Pot-based prescription drug looks for FDA OK (AP)

SAN FRANCISCO ? A quarter-century after the U.S. Food and Drug Administration approved the first prescription drugs based on the main psychoactive ingredient in marijuana, additional medicines derived from or inspired by the cannabis plant itself could soon be making their way to pharmacy shelves, according to drug companies, small biotech firms and university scientists.

A British company, GW Pharma, is in advanced clinical trials for the world's first pharmaceutical developed from raw marijuana instead of synthetic equivalents_ a mouth spray it hopes to market in the U.S. as a treatment for cancer pain. And it hopes to see FDA approval by the end of 2013.

Sativex contains marijuana's two best known components ? delta 9-THC and cannabidiol ? and already has been approved in Canada, New Zealand and eight European countries for a different usage, relieving muscle spasms associated with multiple sclerosis.

FDA approval would represent an important milestone in the nation's often uneasy relationship with marijuana, which 16 states and the District of Columbia already allow residents to use legally with doctors' recommendations. The U.S. Drug Enforcement Administration categorizes pot as a dangerous drug with no medical value, but the availability of a chemically similar prescription drug could increase pressure on the federal government to revisit its position and encourage other drug companies to follow in GW Pharma's footsteps.

"There is a real disconnect between what the public seems to be demanding and what the states have pushed for and what the market is providing," said Aron Lichtman, a Virginia Commonwealth University pharmacology professor and president of the International Cannabinoid Research Society. "It seems to me a company with a great deal of vision would say, `If there is this demand and need, we could develop a drug that will help people and we will make a lot of money.'"

Possessing marijuana still is illegal in the United Kingdom, but about a decade ago GW Pharma's founder, Dr. Geoffrey Guy, received permission to grow it to develop a prescription drug. Guy proposed the idea at a scientific conference that heard anecdotal evidence that pot provides relief to multiple sclerosis patients, and the British government welcomed it as a potential way "to draw a clear line between recreational and medicinal use," company spokesman Mark Rogerson said.

In addition to exploring new applications for Sativex, the company is developing drugs with different cannabis formulations.

"We were the first ones to charge forward and a lot of people were watching to see what happened to us," Rogerson said. "I think we are clearly past that stage."

In 1985, the FDA approved two drug capsules containing synthetic THC, Marinol and Cesamet, to ease side-effects of chemotherapy in cancer patients. The agency eventually allowed Marinol to be prescribed to stimulate the appetites of AIDS patients. The drug's patent expired last year, and other U.S. companies have been developing formulations that could be administered through dissolving pills, creams and skin patches and perhaps be used for other ailments.

Doctors and multiple sclerosis patients are cautiously optimistic about Sativex. The National Multiple Sclerosis Society has not endorsed marijuana use by patients, but the organization is sponsoring a study by a University of California, Davis neurologist to determine how smoking marijuana compares to Marinol in addressing painful muscle spasms.

"The cannabinoids and marijuana will, eventually, likely be part of the clinician's armamentarium, if they are shown to be clinically beneficial," said Timothy Coetzee, the society's chief research officer. "The big unknown in my mind is whether they are clearly beneficial."

Opponents and supporters of crude marijuana's effectiveness generally agree that more research is needed. And marijuana advocates fear that the government will use any new prescription products to justify a continued prohibition on marijuana use. .

"To the extent that companies can produce effective medication that utilizes the components of the plant, that's great. But that should not be the exclusive access for people who want to be able to use medical marijuana," Americans for Safe Access spokesman Kris Hermes said. "That's the race against time, in terms of how quickly can we put pressure on the federal government to recognize the plant has medical use versus the government coming out with the magic bullet pharmaceutical pill."

Interest in new and better marijuana-based medicines has been building since the discovery in the late 1980s and 1990s that mammals have receptors in their central nervous systems, several organs and immune systems for the chemicals in botanical cannabis and that their bodies also produce natural cannabinoids that work on the same receptors.

One of the first drugs to build on those breakthroughs was an anti-obesity medication that blocked the same chemical receptors that trigger the munchies in pot smokers. Under the name Acomplia, it was approved throughout Europe and heralded as a possible new treatment for smoking cessation and metabolic disorders that can lead to heart attacks.

The FDA was reviewing its safety as a diet drug when follow-up studies showed that people taking the drug were at heightened risk of suicide and other psychiatric disorders. French manufacturer Sanofi-Aventis, pulled it from the market in late 2008.

Given that drug companies already were reluctant "to touch anything that is THC-like with a 10-foot- pole," the setback had a chilling effect on cannabinoid drug development, according to Lichtman.

"Big companies like Merck and Pfizer were developing their own versions (of Acomplia), so all of those programs they spent millions and millions on just went away..." he said.

But scientists and drug companies that are exploring pot's promise predict the path will ultimately be successful, if long and littered with setbacks.

One is Alexandros Makriyannis, director of the Center for Drug Discovery at Northeastern University and founder of a small Boston company that hopes to market synthetic pain products that are chemically unrelated to marijuana, but work similarly on the body or inhibit the cannabinoid receptors. He also has been working on a compound that functions like the failed Acomplia but without the depressive effects.

"I think within five to 10 years, we should get something," Makriyannis said.

Italy: Divers find woman's body in stricken ship (AP)

ROME ? Italian Coast Guard divers have found a woman's body in a corridor of a submerged section of the capsized Costa Concordia, raising to at least 12 the number of dead in the cruise liner accident.

Coast Guard Cmdr. Cosimo Nicastro told The Associated Press that the body, wearing a life jacket, was found in a narrow corridor near an evacuation staging point at the ship's rear Saturday.

The body was brought to Giglio, the Tuscan island where the cruise liner hit a reef and ran aground on Jan. 14. Twenty people are missing.

Djokovic routs birthday boy Mahut 6-0, 6-1, 6-1 (AP)

MELBOURNE, Australia ? Novak Djokovic gave Nicolas Mahut one lousy birthday present.

The top-seeded Djokovic routed the Frenchman 6-0, 6-1, 6-1 on Saturday to advance to the fourth round of the Australian Open. He needed only 1 hour, 14 minutes to dismiss the newly 30-years-old Mahut, who lost the longest match in Grand Slam history over 11 hours, 5 minutes against John Isner at Wimbledon in 2010.

"I wish him happy birthday and hopefully tonight he can enjoy it," Djokovic said.

The defending champion is aiming to become the fifth man in the Open era to win three consecutive major titles. He will play the winner of a later match between Milos Raonic and Lleyton Hewitt in the fourth round.

Jo-Wilfried Tsonga, whom Djokovic beat at Melbourne Park for his first Grand Slam title in 2008, also hardly broke sweat in beating Frederico Gil of Portugal 6-2, 6-2, 6-2.

On the women's side, two Wimbledon winners ? Petra Kvitova and Maria Sharapova ? advanced, but two top 10 players went out.

Seventh-seeded Vera Zvonareva was beaten 7-6 (7), 6-1 by fellow Russian Ekaterina Makerova. No. 9 Marion Bartoli lost 6-3, 6-3 to Zheng Jie of China, a former Australian Open semifinalist.

Sharapova was tested for the first time and still came out with a 6-1, 6-2 win over Germany's Angelique Kerber.

The 2008 champion won her first two matches 6-0, 6-1 and has lost five games in reaching the fourth round, but though the scoreline on Saturday made it look easy enough, Sharapova was given a full workout in a 56-minute second set, with many games going to deuce.

After clinching the victory with a forehand winner, Sharapova showed her relief by clenching her fist and screeching "come on!"

"She certainly stepped up in the second set," Sharapova said. "She reached the semifinals at the U.S. Open last year so she's been on the big stage before and I knew she could produce some really good tennis."

She next plays No. 14 Sabine Lisicki, who beat two-time major winner Svetlana Kuznetsova 2-6, 6-4, 6-2.

Sharapova and Kvitova both have a chance of claiming the No. 1 ranking at the end of the tournament. They could play each other in the semifinals, although Kvitova insisted she hasn't looked that far ahead.

"I don't know who lost and who win," the Wimbledon champion said. "No, really, for me doesn't care."

Kvitova reached the Round of 16 when Maria Kirilenko retired with a left thigh injury while trailing 6-0, 1-0 after 38 minutes of their third-round match.

Kvitova next faces Ana Ivanovic, who beat unseeded American Vania King 6-3, 6-4 to reach the fourth round for the first time since she reached the final here in 2008.

That was the same year the 24-year-old Serb won the French Open for her only Grand Slam title, and also claimed the top ranking.

"I still believe I can do well," Ivanovic said. "I'm just enjoying competing again. It's going to be tough (against Kvitova), but I love challenges."

King's loss left Serena Williams as the only American player left in the singles draws after John Isner's loss on Friday ended any hope of a men's champion from the United States.

Five-time champion Williams, hoping to win her 17th straight match at Melbourne Park, was up against Greta Arn of Hungary in a later match Saturday.

Djokovic was ruthless against Mahut, whose movement was hampered by a left leg injury. When the Frenchman won his first game early in the second set, he stretched his arms out to welcome a huge cheer from the crowd as he walked back to the service line.

Djokovic hardly made a mistake as he won 11 of the next 12 games and finished with eight unforced errors. He has won 24 straight sets at the Australian Open, and has lost 10 games in his first three matches this time.

"I always played well in Australia. This is the only Grand Slam I won twice," he said. "The reason that I skipped the opening week of the season was because I wanted to get ready for Melbourne.

"The conditions are great. They're very suitable to my style of the game, day and night. I'm really looking forward to next week."

Like Djokovic, No. 4-ranked Andy Murray was also up against French opposition later Saturday in the shape of Michael Llodra.

In all, six Frenchmen reached the third round. Tsonga wasted hardly any time becoming the first of them to move into the Round of 16. The 2008 finalist took six of his seven break-point opportunities and saved all five against him.

"I had to take it seriously and this is what I did. I'm just really happy to go through," said Tsonga, who will face fellow Frenchman Julien Benneteau or Kei Nishikori of Japan next.

Richard Gasquet, the fourth highest-ranked Frenchman at No. 18, knocked out ninth-seeded Janko Tipsarevic 6-3, 6-3, 6-1. He will play fifth-seeded David Ferrer, who lost the first four games against Juan Ignacio Chela before recovering to win 7-5, 6-2, 6-1.

On the other half the draw, No. 2 Rafael Nadal had a 6-2, 6-4, 6-2 win over Lukas Lacko on Friday and Roger Federer had a 7-6 (6), 7-5, 6-3 win over 6-foot-10 Croatian Ivo Karlovic. Federer will play Australian teenager Bernard Tomic on Sunday.

An early rematch of last year's women's final is already in place, with defending champion Kim Clijsters and Li Na both winning Friday night to set up a meeting in the fourth round.

Clijsters advanced with a 6-3, 6-2 win over Daniela Hantuchova. Li didn't even finish four games before Anabel Medina Garrigues quit with a badly sprained right ankle.

The winner of the Clijsters-Li match will likely face a quarterfinal against top-seeded Caroline Wozniacki, who has not dropped a set in advancing to the fourth round as she continues her quest for a first Grand Slam title. She beat Monica Niculescu of Romania 6-2, 6-2, while third-seeded Victoria Azarenka defeated Mona Barthel 6-2, 6-4.

Wozniacki will next play former No. 1 Jelena Jankovic, who beat Christina McHale of the U.S. 6-2, 6-0.

John King On Newt Gingrich Attack: Question Choice Was 'My Decision, And Mine Alone'

Newt Gingrich's fiery response to CNN moderator John King's question about his ex-wife was the electrifying highlight of Thursday's Republican debate. After the session was over, King defended his decision to ask the question that had everybody talking.

Gingrich flatly condemned King, along with CNN, for asking a question about Marianne Gingrich, who told ABC News earlier on Thursday that her ex-husband had asked her for an open marriage while he was having an affair with his eventual third wife. Gingrich said he was "appalled" by King's decision to lead off the debate with a question about his former wife's allegations, calling it "close to despicable."

Speaking on a CNN panel, King said he had a friendly conversation with Gingrich after the debate was over. He called it a "damned if do you, damned if you don't" situation.

"Is it an issue I'm happy came up?" he said. "Of course not." But he came firmly down on the idea that, since people were talking about Gingrich's past, the issue was worth raising.

"I understood that if I asked the question he was not going to be happy with it, and he was going to turn on me," he said of Gingrich. "...It was my judgment, my decision, and mine alone. If we're going to deal with it, let's deal with it up front."

The other panelists mostly backed King up. David Gergen called Gingrich's infidelity issues "the elephant in the room," and said King had a "duty" to ask the question. Ari Fleischer, George W. Bush's former press secretary, was not so charitable.

"I don't think it was the right sequence," he said, explaining that he thought the question was too geared towards political junkies and not enough towards a broader audience interested in bigger issues.

King parried the disagreement with aplomb. "This is a debatable question," he said. "...I completely understand and respect those who disagree."

Make Windows Explorer's Sidebar Expand As You Navigate Through Folders [Windows Explorer]

Make Windows Explorer's Sidebar Expand As You Navigate Through FoldersA simple option in Windows Explorer's preferences fixes one of its biggest annoyances, automatically expanding the left sidebar as you navigate through your computer's folders.

Perhaps this is an obvious tip, but once I discovered it, I realized how useful it can be. In Windows 7, by default, Explorer's sidebar stays the same unless you click around in it. That is, if you use the right pane to navigate through folders, as most of us do, the left sidebar doesn't change. With a simple checkbox, though, you can have the left sidebar focus on your current folder in the tree as you click around the right pane. To enable this feature:

  • Open up Explorer and go to Organize > Folder and Search Options.
  • On the General tab, look under "Navigation Pane" and check "Automatically expand to current folder".

This is especially useful considering Windows 7's Explorer doesn't have a "one folder up" button. You could always click on the folder's name in Explorer's address bar, too, but this can save you a few clicks if you're constantly jumping around nearby folders. Again, it certainly isn't a new tip, but we've never featured it before, and having just discovered it, I thought it was a pretty cool feature.
